The process was developed for high bacterial biomass production (Shake flask to fermentor), which was used for expressing Transaminase and assay methods for quantitative and qualitative analysis of the enzyme.
•
Isolation and partial purification of Aceto Hydroxy Acid Synthase enzyme from bacteria for the production of chiral drug intermediates.
•
Yeast-based production of phenyl acetyl carbinol (PAC) production using biotransformation of benzaldehyde and acetaldehyde in the bioreactor.
Enzymes and extracellular metabolite studies:
•
Isolation of crude enzymes from bacteria and yeast cells, and developing assays for enzymes like Pyruvate Decarboxylase, Invertase, Alcohol dehydrogenase, Aceto Hydroxy Acid Synthase (AHAS), and Transaminase.
•
Enzyme kinetics: Determination of Km and Vmax values of Pyruvate decarboxylase for pyruvate & benzaldehyde substrates.--

1.Hyaluronic acid (HA) production from bacterial fermentation (Streptococcus zooepidemicus)
•Developed strains to produce low molecular weight Hyaluronic Acid using various mutagenic procedures (UV, NTG, and EMS).
•Optimized nutritional and physical parameters for the synthesis of low molecular weight HA.
•Implemented different strategies, such as H2O2 acclimatization, at the fermentor level (3.0L) to enhance low molecular weight HA production.
2.Lactic acid (HA) production from bacterial fermentation (Lactobacillus rhamnosus)
•Cultivated and propagated Lactobacillus cultures from lab inventory and new cultures obtained from culture collection centers (shake flask study).
• Optimized fermentation processes for lactic acid production in fermenters ranging from 1 to 10 liters (achieved 80% carbon efficiency).
3. Biological coal bed methane (Bio-CBM)
•Generated methane biogenically from coal using synthetic consortia and consortium isolated from various environments at the bottle level.
•Conducted coal adaptation studies and large-scale experiments (45L Bio-reactor) to study carbon balance.
•Scaled up Bio-CBM production in 45 L bio-reactor and Core Flood Reactor (CFR) using coal-specific consortia.
4.Microbial culture maintenance
•Standardized protocols for lyophilization of bacterial and yeast cultures using freeze dryer.
•Managed maintenance of existing and newly added cultures in the lab inventory.
5.Additional Responsibilities
•Analyzed samples (GC, GC-MS, HPLC) and compiled data for reports.
• Presented findings in monthly review meetings.
•Authored a blog on the Learnet platform titled "Biohydrogen Production: Strategies to Improve Process Efficiency through Microbial Routes."
